Accelerated evolution: advancing antibody affinity technology

Native protein affinity ligands are the result of millions of years of evolution. Today, an accelerated evolution is required to improve properties of these ligands to meet the fastchanging needs of biologics manufacturing. This poster describes the use of surface plasmon resonance (SPR) in high-throughput screening for improved alkali stability and elution characteristics of next-generation protein affinity ligands, with retained strong affinity for human antibodies. Over 400 modifications to the antibody affinity ligands were investigated to evaluate their effect on key performance characteristics for industrial purification applications.
