Considering flow rates


Maximum productivity in a chromatography unit operation is achieved by using the highest flow rate which still achieves the removal of specified impurities and recovery of the product. However, all phases of the chromatographic run are not impacted by flow rate in the same way and considerable time-savings can be made. Perhaps equilibration, wash and re-generation phases can be run at maximum flow rates, whereas loading and elution might be limited by sample viscosity, capacity or resolution considerations. Even programmed changes in flow rate during sample application and elution have been reported in order to improve throughput whilst maintaining performance.
